OFFER DESCRIPTION

The infection with hepatitis C virus (HCV) is a major risk factor for the development of hepatocellular carcinoma (HCC). Although new therapeutic approaches seem to eradicate HCV, it does not completely eliminate the risk of HCC. At present, there is no effective treatment of HCC apart from heavy surgeries (resection, transplantation) and its incidence is increasing worldwide. The development of HCC is strongly dependent on a cascade of morphogenesis events following the activation of cellular signalling pathways. HCC develops from polarized epithelial cells of the liver that are hepatocytes, which have remarkable plasticity and can thus undergo cycles of de-differentiation, division and differentiation to ensure the regeneration of the liver. Unfortunately, in severe pathological conditions, this cellular plasticity can lead to an irreversible process of de-differentiation of hepatocytes to a status of stem cells and thus support the development of an aggressive cancer that is often resistant to treatment. We have demonstrated the key role of the δ isoform (PI3Kδ) of class I phosphoinositide-3 kinases in cell polarity, extracellular matrix (ECM) assembly in epithelial cells. The aim is to decipher how PI3Kd-dependent signalling pathways control plasticity and the development of HCV induced HCC resistant to treatment. The main techniques involved 3D organoid cultures from cell lines and biopsies from patients with HCV induced HCC, confocal analysis and immunohistochemistry. Epigenetic and transcriptomic and bioinformatic studies will be performed.   The candidate must be familiar with advanced imaging technics and software and should have ability to drive its own research. Background in cell biology and cell signaling is required. Knowledge on HCV and bioinformatics are advantages.
